ABSTRACT

Objective:To observe the immune factors, coagulation and curative effect of modified Shoutaiwan with Si Junzitang combined with dydrogesterone tablets in advanced age patients with early threatened abortion, and to explore its mechanism of action. Method:The 90 advanced age patients with threatened abortion and kidney deficiency and blood stasis syndrome differentiation in traditional Chinese medicine (TCM) were randomly divided into control group and observation group by random number table, with 45 cases in each group. Both groups took oral dydrogesterone tablets for luteal support. The control group additionally received natural vitamin E soft capsules by oral administration, while observation group received modified Shoutaiwan with Si Junzitang. The course of treatment was 10 days in both groups. The clinical efficacy, TCM syndrome score, immune factors and coagulation factors of the two groups were compared before and after treatment. Result:There was no statistically significant difference in TCM symptom scores, immune factors, and coagulation factors between two groups before treatment. After treatment, the scores of TCM syndromes were reduced in both groups (P<0.05), the proportion of helper T lymphocyte (Th), Th/Ts ratio, D-dimer (D-D) level and fibrinogen (FIB) were reduced while prothrombin time (PT) and the ratio of suppressor T lymphocyte (Ts) were increased in observation group (P<0.05). After treatment, the proportion of Th, Th/Ts, D-D, and FIB levels in observation group were lower than those in control group, while PT and the proportion of Ts were higher than those in control group (P<0.05). The proportion of natural killer cells (NK) had no significant change after treatment, also with no significant difference between two groups. The total effective rate was 84.4%(38/45) in observation group, higher than 64.4%(29/45) in control group (χ2=4.398,P<0.05). There was no obvious adverse reaction in both groups during the treatment. Conclusion:Modified Shoutaiwan with Si Junzitang combined with dydrogesterone tablets can improve symptoms and the therapeutic effect for fetal protection by regulating the immune balance and coagulation function in advanced age patients with threatened abortion.

ABSTRACT

Purpose:@#To study the effect of early restrictive fluid resuscitation (EFR) on inflammatory and immune factors in patients with severe pelvic fracture (SPF ).@*Methods:@#A total of 174 SPF patients in the Department of Orthopaedics, the First Affiliated Hospital of Chengdu Medical College from July 2015 to June 2018 were involved in this study and divided into EFR group (n = 87) and control group (n = 87) using the random number table method. Conventional fluid resuscitation (CFR) was performed in control group, and EFR was performed in EFR group. The incidences of acute respiratory distress syndrome (ARDS) and multiple organ dysfunction syndrome (MODS) during rescue, successful rescue rate, blood transfusion volume, fluid input, and resuscitation time were compared between the two groups. The parameters including prothrombin time (PT), hematocrit (HCT), platelet (PLT) and blood lactate (BL) at the 4th hour after fluid resuscitation were recorded. The levels of inflammatory factors (TNF-α, IL-6, CRP) and immune factors (CD3+, CD4+, CD8+, CD4+/CD8+) were compared between the two groups before treatment and 7 days after treatment. The revised acute physiologic and chronic health evaluation system and the sequential organ failure assessment scores were adopted for evaluation before treatment and 7 days after treatment.@*Results:@#The incidences of ARDS and MODS during rescue in EFR group were significantly lower than those in control group (p=0.015 and 0.010 respectively), and the successful rescue rate in EFR group was significantly higher than that in control group (p = 0.011). The blood transfusion volume, fluid input, resuscitation time in EFR group were significantly lower than those in control group (p = 0.016, 0.002 and 0.001 respectively). At the 4th hour after fluid resuscitation, PT and BL in EFR group were significantly lower than those in control group (p = 0.021 and 0.003 respectively), while HCT and PLT in EFR group were significantly higher than those in control group (p = 0.016 and 0.021 respectively). On day 7 after treatment, TNF-α, IL-6, CRP and CD8+ in EFR group were significantly lower than those in control group (p = 0.003, 0.004, 0.007 and 0.003 respectively), while CD3+, CD4+ and CD4+/CD8+in EFR group were significantly higher than those in control group (p = 0.004, 0.000, 0.007 respectively). On day 7 after treatment, the revised acute physiologic and chronic health evaluation (APACHE) system and the sequential organ failure assessment (SOFA) scores in EFR group were significantly lower than those in control group.@*Conclusion:@#EFR can effectively eliminate inflammatory factors, improve immune function, maintain the stability of blood components, reduce the incidences of ARDS and MODS, and elevate the successful rescue rate in patients with SPF.

ABSTRACT

Objective@#To study the effects of Phellodendron amurense on herpes simplex virus 1 (HSV-1) and cytokines, and to explore the mechanism of Phellodendron amurense inhibiting HSV-1 virus through multiple channels.@*Methods@#Viruses were inoculated into medicine treated HeLa cells. The proliferation of virus was observed by fluorescence microscopy. The transcriptional levels of glycoprotein gD and functional protein US1 on the surface of virus envelope were detected by quantitative (q) PCR. After incubating HeLa cells for 24 h, qPCR was used to detect the expression of intrinsic immune factors such as IP-10, IL-12, IFN-gamma and transcription factor NF-kappa B (P65). The expression and nuclear location of NF-kappa B (P65) protein were detected by immunofluorescence.@*Results@#Fluorescence showed that the proliferation of virus decreased significantly at 8 and 40 ng/ml (P<0.01), and the transcription levels of viral protein gD and US1 decreased (P<0.05). After incubation for 24 hours, the transcription levels of IP-10, IL-12 and IFN-gamma in HeLa cells increased significantly (P<0.01). The transcription level of transcription factor NF-kappa B (P65) also increased (P<0.05), and immunofluorescence showed that the nuclear penetration rate of p65 subunit of NF-kappa B (P65) in each group increased (P<0.05).@*Conclusions@#Phellodendron amurense extract can inhibit HSV-1 by inhibiting the transcription of viral functional protein and promoting the expression of cellular immune factors.

ABSTRACT

Objective To investigate the relationship among serum heat shock protein 70 (HSP70), suppressor of Cytokine Signaling-3 ( SOCS-3) and immune factor in pregnant women with hypertension,and to analyze the diagnostic value of the two indicators. Methods Eighty-six pregnant women with hypertension who were treated in the Second People′s Hospital of Yichang from January 2016 to February 2018 were selected,according to the severity of the disease,they were divided into pre-eclampsia (51 cases) and severe pre-eclampsia group (35 cases),another 40 normal pregnant women in the same period were selected as control group. The serum levels of HSP70 and SOCS-3,plasma immunoglobulin A (IgA), immunoglobulin M ( IgM), immunoglobulin G ( IgG), complement C3 and complement C4 levels were detected in each group,the correlation between serum HSP70 and SOCS-3 levels and immune factors were analyzed. ROC curve was used to analyze the predictive value of serum HSP70 and SOCS-3 in hypertensive disorder complicating pregnancy. The area under the ROC curve of HSP70 was 0. 821,and the critical value of the prediction was more than 0. 89 μg/L,the sensitivity of HSP70 to pregnant women with hypertension was 86. 3%,the specificity was 76. 4%,the area under the ROC curve of SOCS-3 was 0. 759,the critical value of the prediction was less than 0. 035 ng/L,and the sensitivity of SOCS-3 to pregnant women with hypertension was 79. 4%,and the specificity was 71. 6%. Conclusion The abnormal increase of serum HSP70 and abnormal decrease of SOCS-3 in pregnant women with hypertension, maternal serum HSP70 and SOCS-3 levels are closely related to immune factors,the serum HSP70 and SOCS-3 may be used as early predictors of hypertensive disorder complicating pregnancy.

Objective@#In this study, the mRNA of interferon (IFN)-α, IFN-γ, Mov10 and TRIM22, which are innate immune factors, was detected in chronic hepatitis B patients with nucleoside analogues multidrug resistance to investigate host innate immune status and explore the host immune molecular mechanism under the nucleos(t)ide analogues multiresistant HBV infection.@*Methods@#Twenty eight nucleos(t)ide analogue resistant patients and 20 healthy individuals in the control group were collected from infection medical clinic department of Heilongjiang provincial hospital. There were 18 cases of multiple drug resistance and 10 cases of cross resistance.Their peripheral blood specimens were collected. The mRNA of IFN-α, IFN-γ, Mov10 and TRIM22, that are the innate immunity factors in peripheral blood, were detected in 28 nucleos(t)ide analogue resistant patients and 20 healthy individuals using RT-PCR and the data were analyzed by statistical mthods.@*Results@#The mRNA levels of Mov10 and TRIM22 mRNA that are innate immune factors in peripheral blood mononuclear cells of multidrug resistant chronic hepatitis B patients were higher than those of control groups with statistically significant differences (P<0.01). TRIM22 mRNA in chronic hepatitis B patients with multidrug resistance showed positive correlation with IFN-α, IFN-γ and Mov10 mRNA; Mov10 mRNA showed positive correlation with TRIM22 and IFN-α mRNA; IFN-α mRNA showed positive correlation with IFN-γ, Mov10 and TRIM22 mRNA. There were no statistically significant differences in the innate immunity factors IFN-α, IFN-γ, Mov10 and TRIM22 mRNA between multidrug resistant group and cross-resistant group.@*Conclusions@#The mRNA levels of Mov10 and TRIM22, that are innate immune factors, were higher in chronic hepatitis B patients with nucleos(t)ide analogues resistance than those of healthy control group. The correlation analysis of mRNA levels between IFN-α, IFN-γ, Mov10 and TRIM22 expression demonstrated that the correlations among them were different under the nucleoside analogues resistant HBV infection.

ABSTRACT

Objective:To investigate the effect of STIM1 on the survival and proliferation of breast cancer cells and its preliminary mechanism analysis.Methods:Normal mammary epithelial cells MCF-10A as control,the expression of STIM1 in MCF7, HCC1569,MDA-MB-231 and BT549 breast cancer cells were detected by Western blot;STIM1 siRNA sequence(STIM1-siRNA group) were transfected into MDA-MB-231 cells,and the negative control group and the blank control group were set up,the protein expression of STIM1 in each group were detected after cells were transfected for 48 h;MTT method was used to detect cell activity cells were transfected for 24 h,48 h and 72 h;cell apoptosis was detected after cells were transfected for 48 h by flow cytometry;the mRNA expression of IL-6 and TNF-α were detected by RT-PCR;the expression of PCNA,Bcl-2,Bax,Caspase3,STAT3 and p-STAT3 protein were detected by Western blot.Results:The expression of STIM1 protein in breast cancer cells was significantly higher than that in MCF-10A cells (P<0.05);the expression of STIM1 protein in MDA-MB-231 cells transfected STIM1-siRNA was significantly lower than the control group(P<0.05);compared with the control group,cell viability in STIM1-siRNA group decreased significantly in cells were transfected for 48 h and 72 h,the apoptosis rate in 48 h was significantly increased,the expression of IL-6 and TNF-α mRNA sig-nificantly decreased,the expression of PCNA,Bcl-2 and p-STAT3 protein were significantly decreased,the expression of Bax and Caspase3 protein increased significantly.Conclusion:STIM1 gene is highly expressed in breast cancer cells.Inhibition of STIM1 expression by RNA interference can down regulate the activity of cancer cells,induce apoptosis,enhance immunity and by down regulation STAT3 signal.

ABSTRACT

Objective To investigate effect of interferon gemma on immune factor and CD69, CD107a in patients with delayed type hypersensitivity by interferon gamma.Methods 76 cases with delayed type hypersensitivity were selected and divided into 2 groups.38 cases in control group were treated conventional therapy, 38 cases in experiment group were treated by interferon gamma.Peripheral blood CD69, CD107a, immune factor, T cell subsets and the treatment efficiency were compared after the treatment.Results Compared with the control group, the serum CD69 and CD107a levels were lower in the experimental group (P<0.05), serum IgG and IgA levels were higher, the serum IgE level was lower (P<0.05), and the CD3 +,CD4 +,CD4 +/CD8 +level was higher, serum CD8 + was lower (P<0.05) .The effective rate of the treatment group was significantly higher than that of the control group (P<0.05) .Conclusion Interferon gamma has good clinical effect in the patients with delayed type hypersensitivity, and can effectively reduce the levels of CD69 and CD107a in the peripheral blood, and regulate the immune function of the body.

ABSTRACT

Objective To investigate effect of pidotimod on TH1/TH2 balance and immune factor in patients with severe trauma.Methods 90 cases with severe chest trauma patients were randomly divided into two groups.45 cases in control group were treated by conventional anti-inflammatory treatment, 45 cases in experimental group on base of control group with pidotimod 800mg /times, 1 times daily.After treatment, the serum levels of IL-4, IFN-γ, IL-2, IL-10, IL-17,IgM, IgA and IgG were compared.Results Compared with before treatment, indexes were improved(P<0.05). Compared with the control group, IL-4 level was lower, IFN-γlevel was higher, IFN-γ/IL-4 was higher(P<0.05).IL-2, IL-10, IL-17 level was higher(P<0.05).IgA, IgM, IgG level was higher(P<0.05).Conclusion Pidotimod can balance the TH1/TH2 in the patients with severe trauma, and adjust the serum levels of IgM, IgA and IgG,can be used in clinical application.

ABSTRACT

82 spastic cerebral palsy and normal children were selected, including 27 younger and 27 older spastic cerebral palsy( observation group) ,and 14 younger and older normal children( control group) . The tumor necrosis factor α( TNF-α) levels were measured in the serum with ELISA. The TNF-α levels in the serum of the younger and older spastic CP groups were higher than those of the control groups ( P<0.01 ) . In observation groups, the TNF-α levels of younger spastic cerebral palsy were higher than those of the older (P<0.01). On the contrary, the normal children in the control group of different ages had no difference of TNF-α. TNF-αexpression was signifi-cantly higher in the serum of the spastic CP group than that in the control group ( P<0.01 ) . TNF-α expressed continuously in vivo of children with spastic cerebral palsy involve subsequent brain injury.

ABSTRACT

Objective To explore the effect of multiple low dose radiation(LDR)on the apoptosis of splenocytes and immune factors in diabetes mellitus(DM)rats.Methods The rats were randomly divided into control,DM and DM + LDR groups.The irradiation doses were 25,50 and 75 mGy,and the irradiated times were 15.At the fourth weekend after the DM rats irradiated,the apoptotie rate and TCRαβ percentage of splenoeytes were detected by flow cytometry,and the content of IL-2 in both serum and supernatant of cultured splenocytes were detected by ELISA.Results Compared with that in the control,the body weight(BW)decreased in the DM and DM + LDR groups,particularly in DM group.The blood glucose(BG)level in the DM + LDR groups was higher than that in the control,but decreased significantly as compared with that in the DM group(P < 0.01).As compared with those in the control,the apoptotic rate in DM + 50 mGy(P < 0.05)and the content of serum IL-2 in DM + 75 mGy group(P < 0.01)all increased significantly,while the content of IL-2 in supernatant of cultured splenocytes decreased significantly in the DM + LDR groups.Compared with those in the DM group,the apoptotic rate and the percentage of TCRαβ in splenocytes in the DM + LDR groups(P < 0.01-P < 0.001)and the content of IL-2 in serum in DM + 50 mGy group(P < 0.01)decreased significantly.Conclusions The multiple LDR could weaken the loss of BW and increase of BG caused by DM,decrease the splenocyte apoptosis induced by DM,and regulate the immune factors.

ABSTRACT

OBJECTIVE:To observe the effects of Tanreqing injection on the immune factors and clinical outcome in patients with ventilator associated pneumonia(VAP).METHODS:155 patients with VAP were randomly divided into trial group and control group.Serum levels of CRP,IL-6 and TNF-? were determined at 0,1,3,5 and 7 days after treatment.The chest X-ray results at 0 and 3 days after treatment were compared.The mechanical ventilation(MV)time and the successful rate of weaning from mechanical ventilation and the mortality rate were recorded.RESULTS:The serum levels of CRP,IL-6 and TNF-? in all 155 VAP patients were significantly higher than normal levels,at 5 days,serum levels of IL-6 and TNF-? in the trial group began to decrease significantly as compared with the control group(P
